With the advent of the post genomic era, proteomics has achieved unprecedented development. Proteomic research aims to reveal the expression law and biological function of all proteins that truly perform life activities. With the proposal of new concepts such as proteomics, proteomics, functional proteomics and structural genomics, proteomics has become an extremely active discipline in the field of biology. Two dimensional electrophoresis (2DE) is one of the three key core technologies in proteome research. Because of its high resolution and sensitivity, two-dimensional electrophoresis has become a basic tool for analyzing complex protein mixtures.
The first electrophoresis of 2DE was isoelectric focusing electrophoresis (Iso-Electric Focusing, IEF). Then, the protein was electrophoretic in second directions by twelve alkyl sulfonate polyacrylamide gel electrophoresis (SDS-PAGE). In IEF, proteins are separated due to different isoelectric points. In SDS-PAGE, proteins with different molecular weights are separated from each other, detected by Coomassie brilliant blue or silver staining, and the results are compared and analyzed by pdquest and other software.
